Our laboratories are interested in studying and manipulating the function of biological molecular machines. The goal of our joint project is to engineer an important representative, the lactose permease (LacY) in such a fashion that it can be controlled and activated by light. The lactose permease of E. coli is a prototypical membrane transport protein, which catalyzes the co-transport of a carbohydrate molecule with a proton. Using a combination of chemical synthesis and classical protein-engineering, we propose to modify LacY with a photoswitch that can control the molecular movements of the protein. This molecular switch will be synthesized in the laboratories of Dirk Trauner in Munich and attached to the protein and functionally studied in the laboratories of Ron Kaback at UCLA.
